What is Aloxi?
The drug is used to prevent nausea and vomiting associated with cancer Chemotherapy
What does Aloxi contain and how does it work?
The active substance is palonosetron. Palonosetron is used as an anti-emetic and anti-nausea medicine. Palonosetron limits the effect of serotonin (another name for serotonin is 5-hydroxytryptamine, in short 5-HT) naturally occurring in the gastrointestinal tract, nervous system and in the platelets of substances with multiple effects that may cause gag reflex. Palonosetron selectively blocks the 5-HT3 serotonin receptors found in the nerve cells of the central and peripheral nervous systems. In this way, it prevents the stimulation of these receptors by serotonin. Cytostatics (chemotherapy) can cause the release of serotonin in the small intestine thus initiating the gag reflex as a consequence of stimulation of the 5-HT3 serotonin 5-HT3 vaginal nerve fibers. Stimulating the vagus nerve can also trigger central excitation leading to vomiting. Palonosetron inhibits the formation of the gag reflex. Administered intravenously. Palonosetron is metabolised in the liver. 40% of the drug is excreted in the urine in unchanged form, about 50% is converted into virtually inactive metabolites.
When to use Aloxi?
The preparation is indicated in adults:
· In the prevention of acute nausea and vomiting caused by anti-cancer chemotherapy with severe vomiting
· In the prevention of nausea and vomiting caused by anti-cancer chemotherapy with moderate vomiting.
When not to use this preparation?
Unfortunately, even if there are indications to use the preparation, it can not always be used. You can not use the preparation if you are allergic (you are hypersensitive) to any of the ingredients.
Take special care with Aloxi?
Certain diseases and other circumstances may constitute a contraindication to use or indication to change the dosage of the preparation. In some situations it may be necessary to carry out specific check-ups.
The preparation may affect the prolongation of intestinal transit time. Patients with a history of constipation or symptoms of subacute intestinal obstruction
should remain under special observation after taking the preparation.
Although it was not found that the preparation caused a clinically significant elongation of the QT in the ECG, care should be taken in the case of people with congenital QT prolongation in ECG and those at higher risk of ECG. This applies in particular to people who have ever been diagnosed (also if the diagnosis was related to family members) QT prolongation, electrolyte imbalance, congestive heart failure
, cardiac dysfunction (arrhythmias, bradyarrhythmias, conduction abnormalities) and people taking medications that may cause elongation QT interval or electrolyte disturbances. If you belong to any of the above before the start of the preparation, inform the attending physician. All deficiencies, especially potassium and Magnesium
, should be topped up before applying the preparation.
Do not use the preparation for the prevention or treatment of nausea and vomiting in the days preceding chemotherapy.

Does this preparation affect the ability to drive vehicles?
No studies on the effects on the ability to drive vehicles or operate machinery / equipment have been performed. The preparation may cause side effects such as dizziness, fatigue, drowsiness or other symptoms that may impair psychomotor performance and the ability to drive vehicles and operate machinery / machines.
Dosage of Aloxi
The preparation is a solution for injection. It is intended for intravenous administration only by qualified medical personnel. The preparation can be used only before administering chemotherapy. If you have any doubts regarding the use of the preparation, consult a physician.
A single dose of 250 μg in a 30-second intravenous bolus injection 30 minutes before the start of chemotherapy.
There is no need to adjust the dose in elderly patients with impaired liver or kidney function.
The product is not intended for use in children or adolescents.
Can Aloxi be used during pregnancy and breastfeeding?
During pregnancy, do not use any medicine without consulting your doctor!
It is very important that before using any medicine during pregnancy or breastfeeding, consult a physician and explain beyond any doubt the potential risks and benefits associated with the use of the drug. If you are pregnant or plan to become pregnant, please inform the doctor prescribing the prescription for this medicine.
There is no clinical experience regarding the use of the preparation in pregnant women. The preparation should not be used during pregnancy unless the doctor considers it absolutely necessary.
Do not use the product during breast-feeding.
Can I use other preparations in parallel?
Tell your doctor about all medicines you have recently taken, including those that are over the counter.
Take particular care if you are using drugs that may affect the prolongation of the QT interval on the ECG (eg some anti-arrhythmic drugs, quinolone derivatives, some antidepressants and antipsychotics, others).
There was no effect of the preparation on the antitumor efficacy of chemotherapeutic agents (cisplatin, cyclophosphamide, cytarabine, doxorubicin
or mitomycin C).
Parallel use of the preparation with: corticosteroid
s, painkillers, antiemetics and antinauseants, antispasmodics or cholinolytics are considered safe.
What side effects can occur?
Like all medicines, Aloxi can also cause side effects, although they will not occur in all patients who use this medicine. Remember that the expected benefits of using the drug are usually greater than the damage resulting from the occurrence of side effects.
Common: headaches, dizziness, constipation, diarrhea. Uncommon: drowsiness, insomnia, anxiety, euphoria, sensory disturbances (paraesthesia, peripheral neuropathies), eye irritation, amblyopia, tinnitus, motion sickness, increased blood Glucose
